Semester II - Paper II - MCC-201 - Microbial Physiology

Duration of Examination: 3 hrs
Max Marks: 100
Credits: 4

Unit I :

Culture techniques - media preparation - preservation of cultures - Aerobic and anaerobic culture techniques. Growth of bacteria: Batch and continuous culture - factors affecting growth - Growth determination - Growth curve. Microbial morphology - wet mount. Hanging drop, staining methods - dyes -. simple - differential and special staining techniques - acid fast stain, spore stain, capsule stain, staining for metachromatic granules. Development of laboratory techniques, pure and mixed cultures.

Unit II :

Photosynthetic bacteria - classification and salient features of cyanobacteria - cell structures and funtio. Fungi and slime molds classification - morphology, growth, reproduction and dimorphism of Rhizopus, Fusarium, Aspergillus, Agaricus, Penicillium, Candida. Eucaryotic algae - classification - growth and reproduction of chiamydomonas, volvox, chiorella and diatoms.

Unit III :

Basic concepts of metabolism - Transport of nutrients by active and passive transport - Assimilatory and dissimulatory pathways - substrate level and oxidative phosphorylation and ATP generation. Respiration, Fermentation, Photosynthesis - pathways of amino acid sythesis and interconversions. Bacterial enzymes - structure - classification - properties - coenzymes - isoenzymes.
